In Ontario, Canada's most densely populated province, 38,000 marijuana applications were handled in the first few hours of Wednesday, while sales reached about 42,000 in nearby Quebec, through shops and the Internet, breaking all expectations.
A shortage of supplies was reported in the provinces of Newfoundland and Saskatchewan, as well as in the Arctic region of Nunavut.
Meanwhile, many online retailers warned, customers expect a delay in shipping up to five days with late work of night packing orders.
Canadian mail workers are also preparing to start work on Monday, more than a year after the suspension of contract talks, which could delay the delivery of online orders.
The move is a political victory for Trudeau, who pledged to legalize cannabis in his campaign in 2015 in an effort to deprive organized crime gangs of drug profits and regulate the production, distribution and consumption of marijuana consumed by millions of Canadians illegally.
